Human Complement C2 ELISA Kit (ab254501) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Complement C2 protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, milk, saliva, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Complement C2 with 175.11 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Complement C2 is a serine protease involved in both the classical and lectin pathways of complement activation. C2 is secreted as a zymogen. Activation of C2 by C1 leads to the proteolysis of C2 into C2a and C2b. Deficiency of C2 can lead to various types of autoimmune diseases.

Function
Component C2 which is part of the classical pathway of the complement system is cleaved by activated factor C1 into two fragments: C2b and C2a. C2a, a serine protease, then combines with complement factor 4b to generate the C3 or C5 convertase. -
Involvement in disease
Defects in C2 are the cause of complement component 2 deficiency (C2D) [MIM:217000]. A deficiency of the complement classical pathway associated with the development of autoimmune disorders, mainly systemic lupus erythematosus. Skin and joint manifestations are common and renal disease is relatively rare. Patients with complement component 2 deficiency are also reported to have recurrent or invasive infections. -

The Complement C2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Complement C2 were measured in duplicates, interpolated from the Complement C2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1: 4,000, plasma (citrate) 1: 2,000, plasma (heparin) 1: 1,000, and plasma (EDTA) 1: 1,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Complement C2 concentration was determined to be 27.00 mg/mL in serum, 15.46 mg/mL in plasma (citrate), 11.89 mg/ml in plasma (heparin), and 12.51 mg/ml in plasma (EDTA).

The mean Complement C2 concentration was determined to be 117.47 ng/mL in HepG2 cell culture supernatant, 117.91 ng/mL in milk, and 22.87 ng/mL in female saliva.
