Human Cathepsin C ELISA Kit (ab277434) is an in-vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Cathepsin C in serum, plasma (collect plasma using citrate and heparin as an anticoagulant. EDTA is not recommended as anticoagulants for use in this assay) and cell culture supernatants.
This assay employs an antibody specific for Human Cathepsin C coated on a 96-well plate. Standards and samples are pipetted into the wells and Human Cathepsin C present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-Human Cathepsin C antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Human Cathepsin C bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Function
Thiol protease. Has dipeptidylpeptidase activity. Active against a broad range of dipeptide substrates composed of both polar and hydrophobic amino acids. Proline cannot occupy the P1 position and arginine cannot occupy the P2 position of the substrate. Can act as both an exopeptidase and endopeptidase. Activates serine proteases such as elastase, cathepsin G and granzymes A and B. Can also activate neuraminidase and factor XIII. -

Ubiquitous. Highly expressed in lung, kidney and placenta. Detected at intermediate levels in colon, small intestine, spleen and pancreas. -
