Potent and selective inhibitor of bromodomains BRD7 and BRD9. -
Biological description
Potent inhibitor of BRD9 (Kd = 14 nM) and BRD7 (Kd = 239 nM) in in vitro ITC assays. In Alpha screen assays it inhibits BRD9 (IC50 = 75 nM) and BRD7 (IC50 = 3.4 μM) but displayed no inhibitory activity towards BET family proteins. Shows no significant off-target pharmacology against a panel of 324 kinases and 55 GPCRs at concentrations less than 5 µM. Inhibits the binding of BRD9 (0.1 μM) and BRD7 (1 μM) to chromatin in cell-based assays. Inhibits AML cell line proliferation and inhibits AML tumor xenograph growth.
