Handling
Wherever possible, you should prepare and use solutions on the same day. However, if you need to make up stock solutions in advance, we recommend that you store the solution as aliquots in tightly sealed vials at -20°C. Generally, these will be useable for up to one week. Before use, and prior to opening the vial we recommend that you allow your product to equilibrate to room temperature for at least 1 hour.
Amyloid β (42-1) human peptide should be initially dissolved at a concentration of 1mg/ml in 100% HFIP (1,1,1,3,3,3-hexafluoro-2-propanol). This solution should be incubated at room temperature for 1 hour, with occasional vortexing at a moderate speed. Next, the solution should be sonicated for 10 minutes in a water bath sonicator. The HFIP/peptide solution should then be dried under a gentle stream of nitrogen gas. 100% DMSO should be used to re-suspend the peptide. This solution should be incubated at room temperature for 12 minutes, with occasional vortexing. The final solution should then be aliquoted into smaller volumes and stored at -80 °C. For a working solution, add 500-1000 µl of D-PBS (depending on the final concentration to be used) to the peptide stock solution and incubate for 2h at room temperature to allow for peptide aggregation.Need more advice on solubility, usage and handling?
