This kit employs a modification of the bicinchoninic acid (BCA) method for the colorimetric detection of protein concentration or total protein determination in a sample.

This protocol details a method for a high range assay when the protein in the sample is expected to be between 125 and 2000 µg/mL and a high/low range assay for a wider variety of samples where the protein concentration is expected to be between 5 and 2000 µg/mL. Also detailed in this protocol is a method of measuring the total amount of protein in cell lysates in order to normalize protein levels for coupled assays where cell number or total protein is of interest. It is important to note that because the BCA reaction with protein does not reach a true end point, color development continues the longer a sample is left to incubate. However, using the incubation method detailed in this protocol, the color development is significantly slowed, allowing for multiple samples to be assayed simultaneously.
