This data was developed using ab207302, the same antibody clone in a different buffer formulation.Immunohistochemical analysis of paraffin-embedded human pancreas tissue labeling SPINK1/P12 with ab207302 at 1/200 dilution, followed by Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/500 dilution. Cytoplasmic staining on human pancreas is observed. Counter stained with Hematoxylin. Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/500 dilution. Perform heat mediated antigen retrieval with Tris/EDTA buffer pH 9.0 before commencing with IHC staining protocol.

All lanes : Anti-SPINK1/P12 antibody [EPR17585-116] (ab207302) at 1/5000 dilution
Lane 1 : Human pancreas lysate
Lane 2 : Human fetal kidney lysate
Lysates/proteins at 20 µg per lane.
Secondary
All lanes : Anti-Rabbit IgG (HRP), specific to the non-reduced form of IgG at 1/10000 dilution
Predicted band size: 8 kDa
Observed band size: 11 kDa why is the actual band size different from the predicted? This data was developed using ab207302, the same antibody clone in a different buffer formulation.
Blocking and dilution buffer: 5% NFDM/TBST.
Exposure time: Lane 1: 1 minute; Lane 2: 3 minutes.
SPINK1/P12 is a pancreatic secretory trypsin inhibitor, uniquely expressed in pancreas. Human fetal kidney lysate is used as negative control.
SPINK1/P12 contains a signal peptide, thus observed with a MW of 11 kDa (PMCID: PMC2765331).
