Function
DNA-binding component of the FA core complex involved in DNA damage repair and genome maintenance. Recruited to forks stalled by DNA interstrand cross-links, and required for cellular resistance to such lesions. Component of the heterotetrameric CENP-T-W-S-X complex that binds and supercoils DNA, and plays an important role in kinetochore assembly. Component of the APITD1/CENPS complex that is essential for the stable assembly of the outer kinetochore. Plays an important role in mitotic progression and chromosome segregation. -
Sequence similarities
Belongs to the CENPX family. -
Cellular localization
Nucleus. Chromosome > centromere. Chromosome > centromere > kinetochore. Constitutively localizes to centromeres throughout the cell cycle, and to kinetochores during mitosis. - Information by UniProt

All lanes : Anti-MHF2 antibody (ab104236) at 1 µg/ml
Lane 1 : Human liver tissue lysate - total protein (ab29889)
Lane 2 : HEK293 (Human embryonic kidney cell line) Whole Cell Lysate
Lane 3 : U2OS (Human osteosarcoma cell line) Whole Cell Lysate
Lane 4 : HepG2 (Human hepatocellular liver carcinoma cell line) Whole Cell Lysate
Lysates/proteins at 10 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/10000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 9 kDa
Observed band size: 11 kDa why is the actual band size different from the predicted?
Additional bands at: 160 kDa, 46 kDa, 51 kDa, 58 kDa. We are unsure as to the identity of these extra bands.
Exposure time: 2 minutes
ab104236 detects a band at 11 kDa. While this differs to its predicted molecular weight of 9 kDa, the expression profile observed is consistent with what has been described in the literature PMID:20347429. This blot was produced using a 4-12% Bis-tris gel under the MES buffer system. The gel was run at 200V for 35 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with ab104236 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.
