Function
DNA-binding protein that binds to the 5'GGGAATRCC-3' Ikaros-binding sequence. Transcriptional repressor. Interacts with SPI1 and MITF to repress transcription of the CTSK and ACP5 promoters via recruitment of corepressors SIN3A and CTBP2. May be involved in the development of central and peripheral nervous systems. -
Tissue specificity
Highly expressed in skeletal muscle, low levels of expression in heart, thymus, kidney, liver, and spleen. Expressed in the hematopoietic cell lines MOLT-4, NALM-6 and K562. Highly expressed in THP-1 and Mo7e-IL3 cell lines, which have characteristics of myeloid and early megakaryocytic cells respectively. -
Sequence similarities
Belongs to the Ikaros C2H2-type zinc-finger protein family.
Contains 6 C2H2-type zinc fingers. -
Domain
The N-terminal zinc fingers are involved in sequence-specific DNA binding and heterotypic associations with other family members.
C-terminal zinc fingers mediate homodimerization. -

All lanes : Anti-IKZF4 antibody (ab155754) at 1 µg/ml
Lane 1 : Brain (Mouse) Tissue Lysate
Lane 2 : Brain (Rat) Tissue Lysate
Lane 3 : Liver (Mouse) Tissue Lysate
Lysates/proteins at 10 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/50000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 64 kDa
Observed band size: 70 kDa why is the actual band size different from the predicted?
Additional bands at: 200 kDa (possible non-specific binding), 42 kDa (possible non-specific binding)
Exposure time: 16 minutesPlease note that Mouse Liver tissue lysate in lane 3 represents a negative control for IKZF4.
This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab155754 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution ab133406
