ab23657 recognises a band of 105 kDa in NIH 3T3 cell lysates, and 103 kDa in human adult and fetal heart tissue lysates (see image). However, ab23657 does not appear to recognise HDAC7 in human thymus or CHO-K1 cell lysates, which may be due to low expression in these cells (data not shown). -

Function
Responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4). Histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Histone deacetylases act via the formation of large multiprotein complexes. Involved in muscle maturation by repressing transcription of myocyte enhancer factors such as MEF2A, MEF2B and MEF2C. During muscle differentiation, it shuttles into the cytoplasm, allowing the expression of myocyte enhancer factors (By similarity). May be involved in Epstein-Barr virus (EBV) latency, possibly by repressing the viral BZLF1 gene. -

Additional bands at: 55 kDa, 70 kDa, 80 kDa, 85 kDa. We are unsure as to the identity of these extra bands.
Exposure time: 30 secondsThis blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with ab23657 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.
This antibody was raised against an immunogen that is predicted to recognize multiple isoforms of HDAC7.
