General notes
Reproducibility is key to advancing scientific discovery and accelerating scientists’ next breakthrough.
Abcam is leading the way with our range of recombinant antibodies, knockout-validated antibodies and knockout cell lines, all of which support improved reproducibility.
We are also planning to innovate the way in which we present recommended applications and species on our product datasheets, so that only applications & species that have been tested in our own labs, our suppliers or by selected trusted collaborators are covered by our Abpromise™ guarantee.
In preparation for this, we have started to update the applications & species that this product is Abpromise guaranteed for.
We are also updating the applications & species that this product has been “predicted to work with,” however this information is not covered by our Abpromise guarantee.
Applications & species from publications and Abreviews that have not been tested in our own labs or in those of our suppliers are not covered by the Abpromise guarantee.

Relevance
Defects in Bile salt activated lipase (CEL) are a cause of maturity onset diabetes of the young type 8 with exocrine dysfunction (MODY8) [MIM:609812], also known as diabetes and pancreatic exocrine dysfunction (DPED). MODY [MIM:606391] is an autosomal dominant form of diabetes mellitus. The pancreas serves both endocrine and exocrine functions. The endocrine cells are found in the islets of Langerhans. They synthesize insulin and other hormones, and are involved in the pathogenesis of diabetes mellitus. The exocrine cells produce bicarbonate and digestive enzymes and are involved in the pathogenesis of pancreatic malabsorption. The localization of the islets within exocrine pancreatic tissue is suggestive of an interdependency and cross talk between these two cell populations in their normal and in their abnormal function. -

All lanes : Anti-Bile salt-activated lipase antibody (ab87431) at 1 µg/ml
Lane 1 : Human pancreas tissue lysate - total protein (ab29816)
Lane 2 : Pancreas (Mouse) Tissue Lysate
Lysates/proteins at 10 µg per lane.
Secondary
All lanes : Goat polyclonal to Rabbit IgG - H&L - Pre-Adsorbed (HRP) at 1/3000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 78 kDa
Observed band size: 74 kDa why is the actual band size different from the predicted?
Additional bands at: 72 kDa (possible cleavage fragment)
Exposure time: 30 seconds
We hypothesise that the 74 kDa band represents the short isoform of bile salt activated lipase, and the 72 kDa band represents this protein without its signal sequence. -

The same batches of ab87431 detect bands at different molecular weights in different batches of human pancreas tissue lysate. When we tested ab87431 against a previous batch of human pancreas tissue lysate, we observed bands around 74-kDa. However, when the same antibody is tested against our current batch of this lysate, we see bands around 100-kDa. IHC image of Bile salt-activated lipase staining in mouse pancreas formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab87431, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
